I thought of Isla Fisher also when I heard about this. But I will have to go with Christina Hendricks. I liked her on her brief stint on Firefly.

I would love to see Hugh Laurie as either the Professor or Mr. Howwell.

I think you’re off on most, but the right track.

For Gilligan I’d go with an Office co-star, John Krasinski.
I agree with Richter for Skipper.

Even though it’s a stretch, I’d love to see (on a farce) Abe Vigoda and Betty White as the Millionaire and his wife.

For movie star I’d go with current Cover Girl makeup model Drew Barrymore. Yeah I know that’s cheesy, but so is Gilligan’s Island.

For the professor I’d go with Larry the Cable guy… No, seriously I’d go with Neil deGrasse Tyson…

And for Mary Ann? Someone predictable from the Brittany/Jamie Lynn/Paris/Nicole line up.

The professor and Mary Ann would not be given much pressure in this scenario, of course. Tyson would bewilder his costars with real science that everyone has to Google, and the Spears-Hilton type would be coddled through as the unknowing foil.
